Recent experiments demonstrated that iNKT cells can develop into functionally distinct subsets and that the relative abundance of the various subsets is directly responsible for altering several immunological parameters, such as the development of “conventional” CD8 T cells as well as the secretion of chemokines by dendritic cells. However, the signals involved in determining the developmental fate of iNKT cell precursors remain incompletely understood. Using high-dimensional flow-cytometry analysis to delineate iNKT cell development, we establish a function for the T-cell receptor signaling machinery in influencing the development of the various iNKT cell subsets.
